Alteração no potencial osmótico e teor de carboidratos solúveis em plantas jovens de lobeira (Solanum lycocarpum St.-Hil.) em resposta ao estresse hídrico Rev. Bras. Bot.
CHAVES FILHO,JALES TEIXEIRA; STACCIARINI-SERAPHIN,ELIANE.

Anatomical and physiological modifications of micropropagated 'Caipira' banana plants under natural lighy Scientia Agricola
Costa,Frederico Henrique da Silva; Pasqual,Moacir; Pereira,Jonny Everson Scherwinski; Castro,Evaristo Mauro de.
Research about the use of natural light associated to changes in sucrose levels demonstrated potential in promoting in vitro hardiness of tropical climate species, as well as reducing production costs. However, little is known about physiological and structural changes that happen in the process. This study evaluated the physiological and anatomic performance, and ex vitro survival of micropropagated banana plants in response to cultivation conditions, in the stage of in vitro rooting. Shoots of the 'Caipira' cultivar were cultivated in MS medium, supplemented with 1 mg L-1 NAA and 6 g L-1 agar, in which the following treatments were applied: two sucrose concentrations (15 g L-1 or 30 g L-1) and two cultivation conditions (Natural light - greenhouse and...

Assessment of biochemical methods to detect enzymatic depolymerization of polysaccharides ArchiMer
Rigouin, Coraline; Delbarre Ladrat, Christine; Sinquin, Corinne; Colliec-jouault, Sylvia; Dion, M.
To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The range of concentrations of assorted enzymes allowed us to identify the most sensitive and appropriate method to detect polysaccharide degradation. Reducing sugars assays are quantitative, sensitive and almost usable with all kind of polysaccharide but some compounds may interfere with them. When polysaccharide is...

Chemical composition of corn and sorghum grains cultivated in Oxisol with different application methods and doses of zinc Ciencia e Investigación Agraria
Puga,Aline P; de Mello Prado,Renato; Mattiuz,Ben-Hur; Wyllyam do Vale,Diego; Fonseca,Ivana M.
In general, tropical soils present low concentrations of zinc (Zn), and the deficiency of Zn is recognized as a world nutritional problem for cereal production and human beings. Therefore, the main goal of this study was to assess the effects of different methods of Zn application on the quality of corn and sorghum grains grown in Oxisol. Two experiments were set up in the experimental area of UNESP (campus of Jaboticabal, Brazil). The following nine treatments were applied: three doses of Zn by banded application (seed furrows), three doses of Zn by incorporation into soil (0-20 cm depth), foliar application, seed application, and control (no Zn applied). The treatments were arranged in randomized blocks with four replicates. The contents of Zn,...

Effect of carbon source on morphology and histodifferentiation of Araucaria angustifolia embryogenic cultures BABT
Steiner,Neusa; Vieira,Felipe do Nascimento; Maldonado,Sara; Guerra,Miguel Pedro.
The aim of the present work was to establish in vitro conditions for the induction, stabilization and proliferation of embryogenic cultures of A. angustifolia. Pre-cotyledonary staged zygotic embryos inoculated BM medium supplemented with 5 µM 2,4- D, 2 µM BAP and Kin, and 3% maltose or sucrose resulted in 66.7% induction rate. The rate of induction of embryogenic cultures was affected by the carbon source, as well the multiplication and morphology of the embryogenic cultures. Embryogenic cultures maintained in BM medium with maltose presented bipolar morphology. Globular somatic embryos were obtained BM medium with 9% (PEG) and (9%) maltose. These results could establish an in vitro regenerative protocol towards the conservation and improvement of this...

Effect of P depletion on the functional pools of diatom carbohydrates, and their utilization by bacterial communities ArchiMer
Labry, Claire; Delmas, Daniel; Moriceau, Brivaela; Gallinari, Morgane; Quere, Julien; Youenou, Agnes.
Phosphorus (P) limitation of phytoplankton growth is known to affect the accumulation and release of carbohydrates (CHO) by micro-algae. However, relatively little is known about the fate of algal exudates, notably their bacterial degradation. The CHO chemical characterization is also not exhaustive, especially in ‘functional’ pools relevant for phytoplankton physiology (particulate reserve [R] or structural [S] CHO) and for bacterial degradation (dissolved mono- [MDCHO] and polysaccharides [P-DCHO]). In this study, we investigated how P depletion and repletion affect the CHO composition in diatom Thalassiosira weissflogii cultures, and the shortterm response of free and diatom-attached bacteria in terms of abundance and potential βglucosidase activity...

Effect of sediment nearness on the metabolic enzyme activity and energy state of the oyster Crassostrea gigas ArchiMer
Le Moullac, Gilles; Fleury, Pierre-gildas; Le Coz, Jean-rene; Moal, Jeanne; Samain, Jean-francois.
This study was designed to assess in situ the effects of sediment nearness on Crassostrea gigas metabolism. One year-old oysters were reared for 5 months (April-August) in plastic bags on metallic frames at 10 and 60 cm off the sediment at the beginning of April. The management of the energetic resources (storage, consumption) was estimated with respect to biochemical changes of proteins, lipids and carbohydrates. In order to evaluate the effect of the sediment nearness at cellular level, the metabolic rate was evaluated by assessing pyruvate kinase (PK) and phosphoenolpyruvate carboxykinase (PEPCK) enzyme activities and electron transport system (ETS) activity. The metabolic pathways were assessed by measuring ATP and intermediary metabolites such as...

Hardouin, Kevin; Bedoux, Gilles; Burlot, Anne-sophie; Donnay-moreno, Claire; Berge, Jean-pascal; Nyvall-collen, Pi; Bourgougnon, Nathalie.
In order to develop innovative methods for the production of bioactive extracts with potential health benefits, enzyme-assisted extraction has been applied for the bioconversion of Ulva armoricana (Ulvales, Ulvophyceae). The extraction yields, chemical composition, the antioxidant and antiviral activities were determined to assess the efficiency of six commercial enzymatic preparations. Endo-protease treatments significantly increased the extraction yields compared to the control. The organic matter, neutral sugar and protein contents were increased in all extracts compared to an extraction with water, up to 2.0-fold, 2.7-fold and 1.75-fold, respectively. The samples prepared by a multiple-mix of glycosyl-hydrolases and an exo-β-1,3(4)-glucanase revealed...

Ferulic acid depletion by cultured soybean seedlings under action of glucose and methionine BABT
Herrig,Vanessa; Silva,Hélio Cândido da; Ferrarese,Maria de Lourdes Lucio; Ferrarese-Filho,Osvaldo.
Cultured soybean seedlings were used to investigate how glucose or methionine influenced depletion of ferulic acid. Three-day-old seedlings were grown in hydroponic solution containing ferulic acid plus glucose or methionine, and the level of the phenolic acid were monitored in the nutrient culture. The results showed that ferulic acid depletion was more rapid in the presence of those compounds. After 6 h, the increase caused by glucose (0.01 and 0.05 mM) was more pronounced than methionine in the same concentrations. On the other hand, methionine (0.1 and 0.2 mM) increased depletion more significantly than glucose. Results suggested that both compounds might to increase the allelopathic effects of ferulic acid in the seedlings.

Honorato,Claucia Aparecida; Almeida,Luciana Cristina de; Nunes,Cleujosí da Silva; Carrilho,Elma Neide Vasconcelos Martins; Moraes,Gilberto.
The objective of this work was to evaluate the effect of pelletized or extruded diets, with different levels of carbohydrate and lipid, on the gastrointestinal transit time (GITT) and its modulation in pacu (Piaractus mesopotamicus). One hundred and eighty pacu juveniles were fed with eight isonitrogenous diets containing two carbohydrate levels (40 and 50%) and two lipid levels (4 and 8%). Four diets were pelletized and four were extruded. Carbohydrate and lipid experimental levels caused no changes to the bolus transit time. However, the bolus permanence time was related to diet processing. Fish fed pelletized diets exhibited the highest gastrointestinal transit time. Regression analysis of bolus behavior for pelletized and extruded diets with 4% lipid...

Zanotti,Rafael Fonsêca; Motta,Ludymila Brandão; Bragatto,Juliano; Labate,Carlos Alberto; Salomão,Antonieta Nassif; Vendrame,Wagner A.; Cuzzuol,Geraldo Rogerio Faustini.
The present study investigated the germination and vigor of Caesalpinia echinata (Brazilwood) seeds stored at negative temperatures. Recently harvested seeds were cryopreserved at -18º or -196ºC and periodically evaluated for germination, seed vigor and carbohydrate composition. The temperatures did not influence the germination percentages or vigor. The germination percentage decreased from 88% in recently harvested seeds to 60% after 730 days of storage. The different temperature and storage times tested did not affect the vigor seed germination as indicated by the measures of plant growth and survival. The different temperatures used did not cause changes in the carbohydrate composition. The tegument cell walls were rich in lignin, arabinose and xylose....

In vitro Dendrobium nobile plant growth and rooting in different sucrose concentrations Horticultura Brasileira
Faria,Ricardo T. de; Rodrigues,Fabiana N.; Oliveira,Luciana do V.R.; Müller,Cláudio.
Sucrose is a very important component in in vitro culture media, serving as a source of carbon and energy. In this paper, the rooting and in vitro growth of Dendrobium nobile Lindl (Orchidaceae) were studied using different sucrose concentrations (0 g L-1; 5 g L-1; 10 g L-1; 20 g L-1; 30 g L-1 and 60 g L-1), in a modified MS medium containing half the regular concentration of macronutrients at pH 5.8. Greater increases in plant height (4.21±0.6 cm) and high seedling multiplication (1:4) were observed in the 60 g L-1 sucrose treatment, even without the addition of plant hormones. Sucrose concentration in the culture medium did not influence in vitro plant rooting.

Intracellular and intercellular compartmentation of carbohydrate metabolism in leaves of temperate gramineae Rev. Bras. Bot.
POLLOCK,CHRISTOPHER J.; FARRAR,JOHN; KOROLEVA,OLGA A.; GALLAGHER,JOE A.; TOMOS,A. DERI.
This review considers the importance of compartmentation in the regulation of carbohydrate metabolism in leaves. We draw particular attention to the role of the vacuole as a site for storage of soluble sugars based on sucrose, and discuss briefly their characteristic metabolism. We also point out inconsistencies between the observed properties of vacuoles and the behaviour in vitro of the enzymes of fructan biosynthesis that do not support the hypothesis that the vacuole is the site of synthesis as well as of storage. We also consider compartmentation of carbohydrate metabolism between different cell types, using mainly our studies on leaves of temperate C3 gramineae. Here we present evidence of significant differences in carbon metabolism between...

Lectin-binding properties of Aeromonas caviae strains BJM
Rocha-de-Souza,Cláudio M.; Hirata-Jr,Raphael; Mattos-Guaraldi,Ana L.; Freitas-Almeida,Angela C.; Andrade,Arnaldo F. B..
The cell surface carbohydrates of four strains of Aeromonas caviae were analyzed by agglutination and lectin-binding assays employing twenty highly purified lectins encompassing all sugar specificities. With the exception of L-fucose and sialic acid, the sugar residues were detected in A. caviae strains. A marked difference, however, in the pattern of cell surface carbohydrates in different A. caviae isolates was observed. Specific receptors for Tritricum vulgaris (WGA), Lycopersicon esculentum (LEL) and Solanum tuberosum (STA) (D-GlcNAc-binding lectins) were found only in ATCC 15468 strain, whereas Euonymus europaeus (EEL, D-Gal-binding lectin) sites were present exclusively in AeQ32 strain, those for Helix pomatia (HPA, D-GalNAc-binding lectin) in AeC398...

Methodological challenges in carbohydrate analyses R. Bras. Zootec.
Hall,Mary Beth.
Carbohydrates can provide up to 80% of the dry matter in animal diets, yet their specific evaluation for research and diet formulation is only now becoming a focus in the animal sciences. Partitioning of dietary carbohydrates for nutritional purposes should reflect differences in digestion and fermentation characteristics and effects on animal performance. Key challenges to designating nutritionally important carbohydrate fractions include classifying the carbohydrates in terms of nutritional characteristics, and selecting analytical methods that describe the desired fraction. The relative lack of information on digestion characteristics of various carbohydrates and their interactions with other fractions in diets means that fractions will not soon be...
